New transcriptomic insights in two RNAi wheat lines with the gliadins strongly down-regulated by two endosperm specific promoters

چکیده

The gluten proteins of wheat grain are responsible for visco-elastic properties flour, but they also trigger the immune-response celiac disease. In this work, two low-gliadin RNA interference (RNAi) lines that differ promoter driving silencing (D-hordein and γ-gliadin promoters D783 D793 lines, respectively), were characterized at transcriptomic, protein fraction levels in grain. Silencing gliadins provides a readjustment fractions affects to non-gluten (NGP), which increased both RNAi lines. Determination by means R5 monoclonal antibody showed strong reduction content Moreover, fructans, an oligosaccharide linked with development non-celiac sensitivity (NCWS) significantly decreased down-regulation impacts other metabolic processes, particularly on carbohydrate metabolism, enzyme regulator activity response stress. Genes transcription factors regulated ABA up-regulated, could suggest implication phytohormone stress observed

Effective shutdown in the expression of celiac disease-related wheat gliadin T-cell epitopes by RNA interference.

Celiac disease (CD) is an enteropathy triggered by the ingestion of gluten proteins from wheat and similar proteins from barley and rye. The inflammatory reaction is controlled by T cells that recognize gluten peptides in the context of human leukocyte antigen (HLA) DQ2 or HLA-DQ8 molecules. The only available treatment for the disease is a lifelong gluten-exclusion diet. We have used RNAi to d...

Revealing new insights into different phosphorus-starving responses between two maize (Zea mays) inbred lines by transcriptomic and proteomic studies

Phosphorus (P) is an essential plant nutrient, and deficiency of P is one of the most important factors restricting maize yield. Therefore, it is necessary to develop a more efficient program of P fertilization and breeding crop varieties with enhanced Pi uptake and use efficiency, which required understanding how plants respond to Pi starvation. To understand how maize plants adapt to P-defici...

Suppression of gliadins results in altered protein body morphology in wheat.

Wheat gluten proteins, gliadins and glutenins, are of great importance in determining the unique biomechanical properties of wheat. Studies have therefore been carried out to determine their pathways and mechanisms of synthesis, folding, and deposition in protein bodies. In the present work, a set of transgenic wheat lines has been studied with strongly suppressed levels of γ-gliadins and/or al...
